Cell Panels

In order to take the genetic background of different people into account, Evercyte has developed human hTERT immortalized cell lines from different donors. This “Pharmacocellomics” approach is especially important in the field of toxicology as individual genetic donor background might also influence the cellular sensitivity to specific substances. Moreover, Evercyte has established cell panels from specific tissues allowing the establishment of donor specific co-cultures.

1. Establishment of 3D cell culture models combining different cell types isolated and immortalized from one tissue biopsie

Human skin cell panels

Fibroblasts, keratinocytes, melanocytes, microvascular endothelial cells and adipose derived mesenchymal stem cells isolated from one tissue sample were immortalized by either introduction of hTERT alone or, as in the case of keratinocytes, by introduction of hTERT in combination with SV40 early region. The immortalized cell lines were characterized in detail for expression of cell type specific markers and functions.

The human skin cell panel 1 includes the following cell lines: HDF/TERT164, NHEK/SVTERT164, NHEM/TERT164, HDMVEC/TERT164, ASC/TERT164
Human skin cell panels from other donors are available as well. All cells can be ordered individually.

Application:

• Establishment of human skin equivalents using different cells isolated from a single donor 

2. Establishment of test systems using panels of immortalized and characterized cell lines from different donors

The human immortalized cell panels include hTERT-immortalized cells derived from different tissues and donors. The immortalized cell lines were characterized in detail for expression of cell type specific markers and functions.

The following cell panels are offered:

•  Human umbilical vein endothelial cell panel
•  Human dermal skin fibroblast cell panel
•  Testing of cell function in consideration of biological differences between cells from different donors (male/female, etc).

3. Getting insights into changes associated with cellular growth and immortalization using cell panels of normal and continuously growing cell lines

Human normal and immortalized human cell panels

The normal and immortalized human cell panels include umbilical vein endothelial cells, fibroblasts, melanocytes, adipose-derived mesenchymal stem cells and the corresponding hTERT immortalized cell lines. Cells from at least two different donors from the respective tissues are available. The cell lines were characterized in detail for expression of cell type specific markers and functions.

Application:

•  Analysis of differences between normal human cells and the corresponding hTERT immortalized cell line
•  Analysis of differences between tissues
 

Human low and high population doubling level umbilical vein endothelial cell panel

The human low and high population doubling level umbilical vein endothelial cell panels include hTERT immortalized HUVEC/TERT2, 3, 4, 5, 6, 7 at early PD and high PD post transduction. The cell lines were characterized in detail for expression of endothelial cell type specific markers and functions.

Application:

•  Analysis of differences between cell lines at different population doubling levels
•  Analysis of the accumulation of genetic mutation during serial passaging
